Blackbeard is one of the emperors of the sea and of course a very powerful opponent for Luffy. He may be one of the last game villains to take down Luffy, and a fight between these two characters is inevitable. Blackbeard is aiming for a piece like Luffy himself. This obviously sets them up for a confrontation towards the end of the story, and it will be extremely interesting to see if Luffy defeats him or not.
Regardless, Luffy must fight him and surpass him if he is to become the Pirate King. Blackbeard certainly won't let him get an easy run in one piece, and that makes this fight very exciting. Right now, Blackbeard is also growing in strength, and Luffy may be close to his level, given the fact that he has two powerful Devil Fruits and is still looking for more to increase his already great power.

Shanks is a character that Luffy must overcome in the final saga. While Luffy is already very strong, he is not on the same level as Shanks, especially in terms of his Haki control. This was confirmed to us directly in the Elbaf arc, where Shanks was said to be stronger than Loki, who himself is stronger than Luffy at this point. No matter which way hockey fans look at it, Shanks is far superior to Luffy, and the Straw Hat Captain must reach a higher level if he is to match and surpass Shanks.
These two characters will soon meet in one piece and, when that happens, Luffy will have to prove to him that he is a high pirate for the red-haired emperor.

The five elders are the most interesting antagonists a piece. Although they were not active in the ongoing events of the story at all, in the final saga, that has changed drastically. The Five Elders have already been seen in action, and they're clearly formidable villains, especially considering their amazing yokai-based abilities, which Oda demonstrated in the Egghead arc of the story.
Saturn, one of the Five Elders, was one of the main antagonists of the Egghead Island arc. Luffy faced him and all the other Elders in this arc, but was unable to defeat any of them. The best Luffy could do was blow up Mars, but even that required the help of Sanji, Bonney, and Franky. Alone, Luffy couldn't even beat an Elder. In the future, Luffy is sure to fight against the Elder, and it's going to be incredibly exciting for One Piece fans to witness. At the same time, it will give Luffy another boost in terms of notoriety and power.

The previous Nick, Joy Boy, was an absolutely incredible character. It should be noted that while Joy Boy is a mysterious figure who seems to transcend time, Luffy also “became” Joy Boy.
