The Hyuga clan has gained a great deal of control over the Byakugan over the years, being the direct descendants of Hamura Otsutsuki. Shikotsumyaku Naruto is one of the most underrated kekei genkai to exist in the world, and it is now mostly extinct after the death of the Kaguya clan. The last person capable of properly wielding this power was Kimimaro, who emerged as the main antagonist in the first part of the Naruto series.
Those with the power of Shikotsumyaku can freely manipulate their bone structure and change things like bone density. Their bones are so hard that they are practically impossible to break. They can freely manipulate their bone structure to create defensive structures and adapt them to their surroundings to such an extent that the entire terrain is changed by the bones. The defensive capabilities of these bones make them harder than steel, and even the most powerful offensive jutsu can find it incredibly difficult to penetrate them. Shikotsumyaku is called the ultimate defense by many in Naruto's world for this reason.
Quintuple Rashomon
Quintuple Rashomon Hashirama is a very unique jutsu used by Senju in his battle against Madara Uchiha. This jutsu creates five very large barriers in front of the user that are so strong that they can even grab a true Tailed Beast Ball and dispel it.
While not the strongest defense, it can essentially tank mini nukes with ease, and any incoming projectile can be countered with its use. Orochimaru used an inferior version of this jutsu in the story, but Hashirama was the only true master of it. It certainly goes on to serve as the best defense offered in the series.

Quite simply, one of the most impressive defensive abilities in the world of Naruto comes through the Uchiha Mangekyo Sharingan, which grants. Power of Susanoo. At its most basic, Susano'o allows the user to create a skeletal structure around themselves. As the Mangekyo Sharingan becomes stronger and its control improves, Susanoo itself can grow dramatically in size.
Once the Mangekyo Sharingan is fully matured, and especially when it transforms into an Eternal Mangekyo Sharingan, reducing the strain on the body, the user can also manifest a complete formation known as Full Body Susanoo. Susanoo has an immense defensive ability that the vast majority of powerful jutsu cannot penetrate. In the Fourth Great Ninja War, Madara Uchiha showed a glimpse of Susanoo's power when he unleashed it in a battle against the Five Kage, and it was clear how helpless they were against it.

Masashi Kishimoto introduced the Six Paths powers towards the end of the Naruto series, and these abilities granted access to some unique powers such as truth-seeking orbs. The truth-seeking orbs were first awakened by Obito Uchiha when he became the Ten-Tails Jinchuuriki. After him, Madara Uchiha also gained access to them, and Naruto Uzumaki also gained them after awakening the Six Paths Senjutsu.
Truth-seeking orbs Each single element is composed of yin, yang, and yin-yang release. With all these powers, the truth-seeking orbs can essentially nullify any and all jutsu. The power of the Rinnegan is immense and provides great offensive capabilities but also immense defensive capabilities. With the Deva Path, the user can essentially control attractive and repulsive forces, and depending on how much chakra is put into repulsion, it can basically act as an impenetrable defense.
