Jiraiya made a valiant effort to defeat the Six Paths of Pain at the latter's home ground in the Hidden Rain Village. Jiraiya used all of his chakra, his best jutsu, and the help of Toads Fukasaku and Shima while using Sage Mode. In the end, it wasn't enough, Jiraiya defeats one of the Pens but defeats the rest. Based on the apparent power level and Jiraiya's pain loss at the start of the pain arc, no sannin could overcome the six paths of pain in a showdown. Orochimaru stands a better chance than his allies, however, given how his overall strategy isn't too different from Penn's own. Orochimaru may lack the Rinnegan, but he has gathered various jutsu types as part of his scientific experiments. He can also revive the dead with Edo Tensei, such as the deceased Hokage, making him loose like the Six Paths.

With such power, Orochimaru is a fair challenge to the Six Paths of Pain in battle, but in the end, he will still lose. Even with his Hokage minions and his ability to avoid harm by shedding his skin, Orochimaru has no answer for Pain's toughest abilities. The Phantom Path's ability to absorb all chakra is a strong counter to Orochimaru's heavy reliance on jutsu, and even the Giant Serpent Manda can't deal with the offensive Asura and Deva Paths of pain.

Some special Kage can survive or overcome the pain, such as adult Naruto Uzumaki, Minato Namikaze, and Kakashi Hatake. The other cages can last a while against Pain but with Gaara being one of them the chances of actually winning are slim. It's true that this jinchuriki has come a long way from his villainous days in the Chunin Exams arc, but he still lacks what it takes to destroy Pain for good.

Gaara's sand-based jutsu is stronger than ever without a tailed beast assisting him, and those sand barriers will be useful against the likes of Pen's Asura Path. The problem is that Penn's other Paths have more unique abilities that Sand can't handle, such as the Human Path's ability to extract intel into someone's head and kill them in the process. Of all the kage, the third hokage is the strongest who could not defeat the six paths of suffering. His predecessor, Hashirama Senju, would easily dull the pain, and even Minato's extreme speed and brilliant intellect could give him victory against the pain, but Hiruzen Sarutobi was not so lucky. He may be a “Professor of Jutsu” as an experienced shinobi who uses a variety of techniques, but jutsu takes more than volume to defeat pain.

Thanks to the Phantom Path's ability to absorb chakra at will, many of Hiruzen's jutsus would fall flat and fail to harm any of the six afflictions on the battlefield. That factor alone could decide this match, Hiruzen has very few backup options to keep himself in the fight. His physical strength is impressive and was even greater in the past, but working those paths together will not favor the Deva and Asura paths.

Danzo Shimura once commanded the Route and came close to becoming the official Sixth Hokage, both positions spoke volumes about his immense power and influence in the Hidden Leaf Village. He also had the necessary techniques to fight against Sasuke Uchiha and come close to victory, but in the end, Danzo was unprepared for Sasuke's genjutsu moves. In terms of fighting the Six Paths of Pain, Danzo can last a while to fight such an enemy, but his time is limited.

Kisame Hoshigaki definitely falls on the brute side of the strength vs. trickery spectrum. It's true that Kisame can negate chakra by consuming it with Samehada, but it's still not enough to win this fight.
